M0783 Biotin-X, SE  This reactive biotin derivative contains an aminohexanoyl spacer group (termed "X") between biotin and the binding site for proteins, nucleic acids, or other biological molecules. This spacer helps to reduce potential quenching effects of fluorophores attached to the analyte molecule, since biotin binds to "deep" binding sites in avidin or streptavidin. This extra linking arm makes it more accessible for binding, and improves a variety of assays employing biotin-avidin or biotin-streptavidin systems. This amine reactive (succinimidyl ester, SE) derivative can be readily coupled to primary or secondary amines of analyte compounds or proteins under mild conditions. Biotin is a commonly used hapten that can be detected by using its strong affinity for the proteins avidin (from egg white) or streptavidin. Many avidin or streptavidin conjugates are available that have been fluorescently labeled or are conjugated to an antibody or other detection reagent. This biotin labeling compound has been used in many such detection schemes for peptides, proteins, organelle components, monoclonal antibody analyses, oligonucleotide and microbead labelings. See references below for further information on the use of biotin labeling.

Texas RedTM Avidin is ideal for flow cytometry applications using instruments equipped with dye lasers. The great affinity of avidin for D-biotin (Ka = 10-15/M) provides for a great number of applications in biochemistry (immunoassays, receptor and histochemical studies, bacteriophage inhibitions). This fluorescently labeled avidin conjugate combines stoichiometrically with biotin, having 4-biotin binding sites per avidin molecule. In addition, the Texas RedTM Avidin is a highly cationic glycoprotein that can selectively bind to a component in human and murine mast cell granules in fixed-cell preparations, and can thus be used to identify mast cells in normal and diseased human tissue without requiring a biotinylated probe. See also our Fluorescein Isothiocyanate (FITC) Avidin Conjugate, Product M1274.

M1274 Avidin Fluorescein Isothiocyanate (FITC) Conjugate  Fluorescein, Texas RedTM as well as TAMRA conjugates of Avidin and Streptavidin have become increasingly useful for DNA and RNA microarray analyses, as a method of detecting biotin-labeled antibodies, or in ELISA assays. FITC Avidin is a highly fluorescent conjugate of Avidin and fluorescein isothiocyanate. This green fluorescent protein excites at 494 nm with an emission maximum at 518 nm. Since the excitation and emission maxima are well separated from those of other fluorophores such as Texas RedTM and coumarin, FITC Avidin can be employed with other conjugates to simultaneously localize two or three antigens in the same tissue section. FITC Avidin is ideal for flow cytometry applications using instruments equipped with dye lasers. In addition, the FITC Avidin is a highly cationic glycoprotein that can selectively bind to a component in human and murine mast cell granules in fixed-cell preparations, and can thus be used to identify mast cells in normal and diseased human tissue without requiring a biotinylated probe. See also our Texas RedTM Avidin Conjugate, Product M1124.
M1282 Ethidium Bromide  Commonly used nucleic acid stain. Fluorescence of Ethidium Bromide increases 25-fold upon intercalation with double-stranded DNA and 21-fold upon intercalation with double-stranded-RNA, eliminating the need to destain background.
M1331 3,3'-Dipropylthiacarbocyanine iodide  DiS-C3(3) is a membrane potential sensitive dye for monitoring changes in the cellular and mitochondrial membrane potential in vivo. This dye can exhibit fluorescence changes of up to 80% upon membrane hyperpolarization (where the inside of the cell becomes more negative). The fluorescence emission and excitation (EX = 556 nm; EM = 575nm in MeOH; EX = 520 nm, EM = 595nm in water) are in the red region of the visible spectrum, allowing co-staining with other viability stains.
M1356 MarkerGeneTM Biotin Detection and Quantitation Kit  The MarkerGeneTM Biotin Detection and Quantitation Kit allows the measurement of the molar ratio of biotin to protein in biotin-protein conjugates, biotin-DNA molar ratios in biotinylated DNA samples, or quantitation of biotin concentrations in solution. The assay employs the sensitive chromogenic reagent 4'-hydroxyazobenzene-2-carboxylic acid (HABA) that exhibits a UV spectral absorbance at 500 nm when bound to avidin. Because biotin easily displaces HABA from HABA/Avidin complexes, the resulting decrease in absorption at 500 nm can be used to sensitively measure biotin concentrations. The kit can be used to determine biotin concentrations down to 1 uM concentrations. The assay can be performed in a microtiterplate format.
