Thioflavin-T (ThT) has become among the most widely used dyes for selectively staining and identifying amyloid fibrils both in vitro and in vivo.
Thioflavin T (ThT) is a fluorescent cell-permeable benzothiazole dye that exhibits a shift and increase in fluorescence with eximer formation upon binding to amyloid protein aggregates. ThT has been used to visualise stacked β sheets in vitro and in vivo. Binding induces a shift in excitation maximum (385 nm to 435 nm) and emission maximum (435 nm to 480 nm). It is highly soluble in water, but has limited ability to cross the blood-brain barrier.
